Release of triticum aestivum Whole Exome Product: Hundreds of Whole Exome Programs Facilitate Precision Breeding

Relying on its self-developed IGT® Oligo Pools synthesis platform and TargetSeq® technology platform,  iGeneTech has launched the "Hundreds of Whole Exome Programs". A variety of whole exome products for animals and plants are being successively released. This article will present to you the excellent performance of the triticum aestivum whole exome product.

As one of the most important food crops in the world, the yield and quality of triticum aestivum are crucial to the food and clothing of billions of people. However, the genome of common triticum aestivum is huge (approximately 14.6 Gb), with a high proportion of repetitive sequences and it is an allopolyploid hexaploid, which poses great challenges to the genetic research of triticum aestivum.

Although whole-genome sequencing data is comprehensive, it generates a huge amount of data, which is difficult to analyze and expensive. In contrast, whole exome sequencing can precisely focus on the exome region. As a crucial part of genes that encodes proteins, exons account for only about 1% of the triticum aestivum genome, but they contain the core genetic information that determines various traits of triticum aestivum, greatly reducing the sequencing cost and the complexity of subsequent analysis. Compared with whole-genome sequencing, it can more efficiently screen out genes related to important traits of triticum aestivum, saving a great deal of time and energy for scientific researchers and fully driving the efficient output of scientific research achievements.

iGeneTech's triticum aestivum whole exome product (TargetSeq® Triticum aestivum Exome Panel) is designed based on the 132.6 Mb CDS region of the IWGSC RefSeq V2.1 reference genome. With 2,554,727 unique probes, it can efficiently and uniformly capture the CDS regions of over 100,000 triticum aestivum genes.


Comprehensive coverage of the CDS region

The TargetSeq® Triticum aestivum Exome Panel is designed to achieve a coverage rate of 98.2% for the CDS region. When validated using different varieties such as Chinese Spring, AK58, and Fielder, the coverage rates are all above 95%. In particular, the coverage rate for Chinese Spring is over 98%, enabling an extremely high coverage of the CDS.

High capture efficiency of the target region

The TargetSeq® Triticum aestivum Exome Panel, when paired with the TargetSeqOne® hybridization capture system, can efficiently enrich the target region. The capture efficiency is over 80%, and the flanking capture efficiency can reach up to 87%, far exceeding that of other manufacturers in the market.

Meanwhile, with a data volume of 10 Gb on the MGI platform, an effective sequencing depth of over 45× can be achieved.

Higher uniformity performance

The TargetSeq® Triticum aestivum Exome Panel demonstrates higher uniformity, with a T 20%× coverage rate (%) of up to 97%. With the same data volume, it can effectively improve the coverage and accuracy of exon detection.
