Gene Capture Uniformity Indicators
iGeneTech has been deeply engaged in the gene capture industry for 11 years. It has always adhered to taking clinical needs as the core and built full-chain service capabilities covering probe design, synthesis, and capture reagents.
All along, iGeneTech has strived for excellence in hybrid capture data indicators, and has pursued the optimal performance particularly for uniformity, a core indicator. This article will break down the technical details and key links to comprehensively demonstrate its solid technical strength.
01 AIExome V5 Breaks Through in Homogeneity
Launched in March 2025, AIExome V5, relying on innovative probe design and optimization, effectively reduces biases in high-GC or low-GC regions, ensuring more uniform coverage of target regions. Its uniformity indicator (Fold 80) has achieved a strong breakthrough, reaching 1.24. Even when matched with the latest 2-hour fast hybrid capture protocol, Fold 80 can still be stably controlled at 1.5, and its performance far surpasses that of competitors' products on the market.

02 Even with 2-hour fast hybridization, it can reach the level of overnight hybridization achieved by competitors' hybrid capture reagents.
The fastest 2-hour fast hybridization system is realized, ensuring high-quality and efficient enrichment of target region fragments within a short time, which significantly improves scientific research efficiency.
03 Stable and Excellent Uniformity of Multiple Hybridization Samples
When matched with TargetSeq® Eco Universal Blocking Oligo, it can block libraries within 6 μg, with excellent performance in capture efficiency and uniformity.
Full-Chain Decoding of the Reasons for Improved Uniformity
01 Ten Years of In-Depth Engagement in Probe Design
With the 3-Layer Shingled Probe Design and thermodynamic evaluation, probe capture is effectively ensured at the design stage, enabling the panel's uniformity indicators to gain an advantage from the "starting line".

02 High-Throughput and High-Quality Synthesis Platform
The Ignite 3.0 Platform, equipped with a 680K high-throughput synthesis chip, delivers excellent uniformity (95/5 percentile ratio = 1.82), ensuring equal sampling of each sequence in downstream screening.

03 Simple and Efficient Hybrid Capture System
Whether in low-GC or high-GC regions, the TargetSeq One® v3.0 Capture Protocol can achieve more uniform capture performance.
04 High Depth Ensures Hotspot Detection in Low-GC Regions
The increased depth in low-GC regions not only brings about an improvement in overall uniformity, but also provides sufficient data support for the hotspot depth in low-GC regions.
Figure A: Heatmap Analysis of Depth Distribution Across Different GC Content Intervals. TargetSeq One® v3.0 achieves better uniformized depth in low-GC regions and more balanced overall depth.
Figure B: In the figure, the uniformized depth of 220 hotspots with GC < 40% (including BRCA1/2, KRAS, and PIK3CA among them) is marked in red. In the v3.0 buffer, only 3 sites (1.4%) have a uniformity depth below 0.5, while in competitors' (buffers), as many as 63 sites (28.6%) have a uniformity depth below 0.5.
